Engineering zinc-finger transcription factors (Cardiff University)

We are currently working on a BBSRC funded project to engineer the DNA binding domain of zinc-finger transcription fingers to switch its binding on/off in response to a small molecule effector. The new proteins are being designed with a small molecule binding site in one of the fingers, and the residues lining this binding site will be optimized using a phage display procedure. This approach allows us to choose the small molecule effector at will to obtain desirable in vivo properties (bioavailability, low toxicity).
